Natural products are an important source of drug discovery, and traditional strategies for direct isolation, in situ activation of synthetic gene clusters, and heterologous expression of natural products have problems such as low research efficiency, high repetition rate, and high work intensity. In order to break through the aforementioned bottlenecks, our research group has introduced the concept of synthetic biology into the field of natural product discovery, focusing on the synthesis genes (clusters) of terpenoid natural products from microorganisms and plants. Using various methods such as AlphaFold2 structure prediction, he has excavated and screened a large number of new biosynthetic genes (clusters), and established corresponding synthetic gene databases; Constructing efficient microbial chassis such as Brewing yeast, Aspergillus oryzae, and Streptomyces, utilizing near source hosts to enhance the adaptability and yield of synthetic gene (cluster) heterologous expression; Translate the conventional manual experimental process to a high-throughput automated platform to achieve high-throughput reconstruction and combinatorial rearrangement of synthetic genes, attempting to change the current situation of low flux, low yield, high repetition rate, and unclear biosynthetic mechanisms in natural product discovery.

Prof. Tiangang Liu has been engaged in metabolic engineering and synthetic biology research for a long time, dedicated to efficient synthesis of microbial natural products and high-throughput genome mining and transformation. He led a research team to tackle the problems of low production efficiency and slow discovery speed of complex natural products, and established an automated microbial casting platform and microbial metabolite detection platform, Developed microbial targeted sequencing methods and analysis methods based on in vitro reconstruction and cell-free systems, created chassis cells for efficient synthesis of various natural compounds, and helped improve the efficiency and level of industrial synthesis of various natural products. Recently, an efficient chassis has been combined with an automated platform for genome mining of natural products, achieving batch mining of new compounds. As the project leader, He presided over the national key research and development plan synthetic biology special project, and as the first complete person he won the top ten scientific and technological events in Hubei in 2018 and the first prize for scientific and technological progress in Hubei Province in 2019. As the corresponding author, He has published hundreds of papers such as Nature, Nature Catalysis, and etc.
